The Best Nootropic You Dont Know About J-147 Dihexa and Noopept Explained
I'm not a nootropic head. I've always said therapeutic testosterone is the best nootropic I've ever used, and getting my hormones dialed in did more for my brain than any pill ever has. But I'm genuinely loving our new product BioMind from BioLongevity Labs. It stacks three compounds that work through completely different pathways, and the synergy is real. Here's what each one does and how I actually use it.
J-147
J-147 is the newest of the three. Most people in the research world have heard of dihexa and noopept, but J-147 is still pretty novel.
It's a synthetic derivative of curcumin, developed at the Salk Institute as a candidate Alzheimer's drug. The big advantage over regular curcumin is bioavailability. J-147 is orally active, crosses the blood brain barrier, and is classified as a neuroprotector for its anti-aging properties.
How it works. J-147 binds to the mitochondrial ATP synthase (ATP5A) and modestly inhibits ATP synthesis, which raises intracellular calcium and activates the CAMKK2/AMPK/mTOR pathway. In plain English, it helps get more energy to your brain. It also upregulates BDNF and NGF, which support neuron growth, survival, and plasticity. Plus it brings the antioxidant and anti-inflammatory effects you'd expect from a curcumin derivative.
The evidence. In aged Alzheimer's mice, J-147 reversed cognitive impairment and rescued memory deficits even when given late in disease progression. That's huge, because Alzheimer's treatment is usually about how early you can catch it. Treated mice showed higher hippocampal BDNF, NGF, and synaptic protein levels. In one trial it was comparable to donepezil for short-term memory in scopolamine-treated mice, and stacking the two worked even better.
J-147 entered Phase 1 human trials in 2019. Detailed data isn't published yet, but it's moving forward.
I'm pretty cognizant of brain health because I almost certainly have CTE from getting concussed for 17 years straight playing football. The neuroprotection against amyloid and tau is something I pay attention to.
Typical dose is 5 to 30 mg per day, with 10 mg being my recommended dose.
Dihexa
Dihexa is an oligopeptide derived from a fragment of angiotensin IV. Researchers at Washington State modified it to be stable, orally bioavailable, and able to cross the blood brain barrier.
The headline claim on dihexa is wild. In cell assays, it's reportedly around 10 million times more potent than BDNF at stimulating new synapse growth. It's been called a procognitive and synaptogenic compound for its ability to repair and grow neural connections.
How it works. Dihexa activates the HGF/c-Met pathway. By binding to hepatocyte growth factor and modulating the c-Met receptor, it powerfully stimulates synaptogenesis and dendritic spine formation. In cultured hippocampal neurons, dihexa increased dendritic spines about threefold, similar to HGF itself, and the new synapses were functional.
The evidence. In rodents, dihexa dramatically improved spatial learning. Cognitively impaired rats treated with dihexa performed indistinguishably from healthy controls in the Morris Water Maze. Treated rats had significantly increased synaptic density and spine head size in the hippocampus.
There aren't published peer-reviewed human trials of dihexa itself yet, but a closely related prodrug called ATH-1017 (fosgonimeton) is in clinical development for Alzheimer's, working through the same HGF mechanism.
Anecdotally, dihexa builds up over time. Unlike a stimulant, the effects accumulate as your brain physically adds more synaptic connections. Most users dose 5 to 40 mg per day. I like 10 mg in this stack. Side effects are minimal, though some people report mild nerve tingling or headache at higher doses.
One thing worth flagging. Because dihexa activates a growth factor pathway, there's a theoretical concern about chronic stimulation and tumor growth. No tumors have been reported in animal studies, and at the doses we're talking about I'm not personally worried, but it's the reason I cycle this product.
Noopept
Noopept is a small dipeptide nootropic developed in Russia at the Zakusov Institute as an analog of the racetam family. It was designed off the structure of piracetam and is roughly 1,000 times more potent.
It has a short half-life of 5 to 10 minutes in rodents, but it produces longer-lasting effects through an active metabolite called cycloprolylglycine (CPG), an endogenous neuropeptide.
How it works. Noopept enhances cholinergic signaling, upregulates BDNF and NGF, modulates glutamate receptors, prevents memory loss from scopolamine, protects neurons from glutamate toxicity, and has anxiolytic effects.
The evidence. Most of the research is Russian and largely preclinical. In rats given beta-amyloid to mimic Alzheimer's, noopept improved memory tasks after 7 days, whether given preventively or as treatment. It blocks amnesia from glutamate and cholinergic blockade. It increases BDNF and NGF in the hippocampus and cortex.
In a small open-label human study, 56 days of noopept at 10 mg twice daily improved MMSE scores in mild cognitive impairment patients from 26 to 29 on average. The control group on piracetam improved less.
Anecdotally, users describe a clear-headed feeling, faster thinking, better focus, and a mild psychostimulant effect without the jitters. Most people find noopept gives a focus and clarity boost within days, while memory benefits build over longer use. Standalone dose is usually 10 to 30 mg per day.
Where each one shines
Memory and learning. All three help, but dihexa is the heavy hitter for long-term memory storage by adding synapses. J-147 reverses age-related deficits. Noopept sharpens encoding and recall faster.
Focus and attention. Noopept wins here. It's the closest to a stimulant feel without the side effects.
Neurogenesis and synaptic plasticity. Dihexa is in a class of its own for synaptogenesis. J-147 is strongly neurotrophic. Noopept supports both more modestly.
Neuroprotection. J-147 was literally screened for neuroprotection. It shields neurons from amyloid, glutamate excitotoxicity, and oxidative stress. Noopept has solid neuroprotective data too. Dihexa protects by repairing.
Why the stack works
Each compound hits a different primary pathway. J-147 works on mitochondrial ATP synthase and broad neurotrophic signaling. Dihexa works on HGF/c-Met synaptogenesis. Noopept works on cholinergic and glutamatergic signaling plus BDNF/NGF.
Stacking them means you cover structural growth, metabolic and trophic support, and neurotransmitter optimization all at once. Dihexa builds new synapses, J-147 keeps the neurons healthy and energized to maintain them, and noopept makes sure the cholinergic signaling is there to actually use those connections.
Because they synergize, you can run lower doses of each and still get strong effects.
How I cycle BioMind
Here's the protocol Alex Cuckle helped formulate, and what I personally do.
BioMind two to three times per week. Two capsules first thing in the morning. Each capsule has 5 mg of each compound, so two capsules gives you 10 mg of each.
I know it's weird to sell a product and tell you not to use it daily, but I genuinely don't think you should run this every day. These compounds modulate powerful growth and neurotransmitter pathways. Used daily, you risk desensitizing receptors, driving too much synaptic growth, or pushing BDNF past optimal. BDNF is a Goldilocks molecule. Too little is bad, too much causes excitability and anxiety.
Pulsing gives the nervous system time to integrate, consolidate, and recalibrate. Same logic as training a muscle. Stimulus then recovery.
Alpha-GPC 600 mg on the off days. Alpha-GPC is a bioavailable choline donor and acetylcholine precursor. On non-BioMind days, you're not pushing plasticity pathways, but you're still supporting neurotransmitter production, maintaining mental clarity, and preventing the choline depletion that can cause brain fog after noopept use.

My take
I steer away from most nootropics because they make me anxious and make my brain run too hot. BioMind doesn't do that. My cognition is sharper, reading and learning feels faster, and I haven't had any of the wired feeling I usually get from this category.

Hey everybody, this is Hunter Williams. I hope you're doing amazing wherever you are in the world. Well, I better bring my A game for this one in terms of my verbal fluency and cognition. So what I'm going to do today is introduce three pretty novel, unique, nootropic compounds that are part of our new product called BioMind from BioLongevity Labs. J 147 dihexa and new pep and those are the three things we have in our Neutropics shout out to Alex kickle for helping us formulate and develop that If
you've watched my channel for some time, you probably know. I'm not the biggest nootropic head I always say therapeutic testosterone is the best Nootropic. So I had a profound change in my cognition and brain horsepower once I got my hormones optimized and started therapeutic testosterone. But I will say I am really, really loving this product. So what I'm going to do today is just walk through each of these ingredients and kind of explain the background on them and explain my best case use of these products and how I use them, how cycle them and all that good stuff.

Without further ado, I'll show my screen and today we're gonna learn about these really cool nootropic compounds. Alright, I'm Hunter Williams and today we're going to learn all about J147, Nupept and Dihexa, which is in a product called BioMind and I am just going
walk through the ingredients of that and how to use it. So first I want to talk about the J 147. This one is more of the novel compound. Most people are familiar with DiHexa and NuPept. Those have been served sold in the research world for quite a bit of time now, but J147 is pretty cool. And I will say having used dihexa and nupept in isolation previously, J 147 itself and synergistically alongside those two really kind of takes it over the edge. So not in a bad way, but just in in good way and kind of helps get the most bang for your buck out of those.
So what is it? It's actually a synthetic derivative of curcumin, which is from turmeric. It was developed by researchers at the Salk Institute as an experimental Alzheimer's drug candidate. is designed to be more potent and bioavailable than curcumin, if you know anything about that world, which I am not an expert in by any means, but a lot of the curicumin slash tumeric curculinoid thing comes down to bio-availability and how much bio availability you can get out of that because there's a It is orally active and crosses the blood brain barrier classified as a Jiro neuro protector for its anti-aging and neuro protective property.
So pretty cool. And again, that is where it has been studied. Let's look at how it works. Unlike traditional Alzheimer drugs, it a pleiotropic mechanism. Its primary molecular target was identified as the mitochondrial alpha F1 ATP synthase, otherwise known as ATP5A. By binding to ATP 5A, J147 modestly inhibits ATP synthesis activity, which increases intracellular calcium levels and activates the CAMKK2 AMPK mTOR pathway
associated with cellular energy metabolism and aging. Basically what that means is it helps get more energy to your brain. So it has neurotrophic factor, so it markedly up regulates neurotrophy factors, including BDNF, which is good for us, to an extent, and NGF which has nerve growth factor in the brain, So this leads to enhanced signaling for neuron growth, survival and plasticity. It also exhibits antioxidant and anti-inflammatory effects, a la the curcumin, extract and even influences peripheral metabolism which activates liver AMPK and reduces fatty acids. So notably it has showed antidepressant like effect in rodents, which I can attest to myself.
Not that I was depressed, but it definitely has more of a mood uplifting effect, possibly via raising serotonin and reducing anxiety behaviors. We look at some of the evidence J147 has demonstrated robust cognitive benefits and neuroprotection of multiple rodent models in aged, transgenic Alzheimer's mice. It reversed cognitive impairment and rescued memory deficits even when given at late disease stages, which is pretty cool because a lot of these things that are treatments for Alzheimer is really how fast you can catch it before it gets too bad. And then treated Alzheimer s mice performed better in memory tasks and showed increased hippocampal BDNF and NGF levels and synaptic proteins important
for learning. In another trial, it was comparable to a drug known as Donapizil in improving short-term memory in scopolamine-treated mice, and combining J47 with the DonaPizl had the best effect, actually. I don't know a lot about that drug, but it seems pretty cool. Other studies reported J147 protects neurons from toxicities associated with Alzheimer's, like the amyloid and tau proteins, which I'm very cognizant of because more likely than not, I have a lotta CTE in my brain from being concussed for 17 years in a row when I played football.
It also improves spatial memory in aged normal mice and even extends lifespan and accelerated aging mouse models. So it's neurotrophic and memory enhancing activities correlate with increased BNF signaling and markers of synaptic plasticity. And as of recent years, J147 has entered human testing. A phase one clinical trial started in 2019 evaluated safety in healthy adults. While detailed human data is not yet published, the successful completion of animal toxicology studies and investigational new drug status mark an important step towards potential therapeutic use.
So when we look at this anecdotally from myself and other people in the community, biohackers who have experimented with it, available as a research compound often report enhanced mental clarity and energy. And typically the dose is going to range from five to 30 milligrams per day with 10 milligrams a day being the common and my recommended dose. However, obviously these ports are anecdotal. Take them with a grain of salt. You should always put what I say and anyone else says to the test for yourself or at least do extensive research on your own. Now let's look at dihexa. Not as new as something like J147, but let' go over it anyway.
It's an oligo peptide derived from the angiotensin 4 hormone fragment. So it was invented by researchers at Washington State. modified angiotensin 4 to create a more stable brain penetrant molecule. So unlike the short-lived angiotensine 4, dihexo is orally bioavailable, crosses the blood-brain barrier, and remains intact longer. This small peptide was originally dubbed a pro-cognitive and synaptogenic compound for its unprecedented ability to repair and grow neural connections. In fact, cell assays of synapsin function, reported to be extraordinarily potent on the order of 10 million times more potent than BDNF at stimulating
new synapse growth. So pretty interesting. Anybody that's used it knows in a dose dependent manner, it's relatively mild, but it does have some good benefits. The key mechanism of its activation or its action is the activation of the HGFC met pathway. It binds with high affinity to the hepatocyte growth factor and acts as a positive modulator of HEF receptor cement. So by potentiating HEf and cement signaling, dihexa powerfully stimulates synaptogenesis and dendritic spine formation in neurons.
In cold-herd hippocampal neurons, the HEXA treatment increased dandritical spines around threefold akin to effect of HGF itself. And these new synapses are functional, expressing normal synoptic proteins. An HGF antagonist blocked dihexas synaptogenic and memory effects, indicating it works by amplifying this endogenous regenerative pathway. And H-G-F slash C-MET signaling plays a role in brain repair and plasticity, especially after injury or in neurodegeneration. Dihexa effectively taps into this whole pathway, so aside from the synoptic growth, di hexa also promotes neurotrophic support.
So it's been described as a neurogenic peptide that can trigger neuron growth and differentiation. So, dihexid has been used in lab protocols to drive stem cells to mature into neural or hepatocyte-like cells, highlighting its growth factor activity. Importantly, Dihexid does not simply mimic angiotensin activity, rather it represents a new class of compound targeting growth factors signaling for cognitive enhancement. So when we look at animal studies, DIHEXID has shown pro-cognitive effects in rodent models. Dihexo dramatically improved spatial learning. In the Morris Water Maze, a high dose of the dihexotreated impaired rats performed indistinguishably from healthy controls.
Even systemic administration was effective. DiHexo was given intraperitoneally or orally. and the rescued maize performance in cognitively impaired rats, showing dose-dependent improvements in memory and retention. In aged rats with natural cognitive decline, oral dihexa also improved learning and memory on water maze tasks, although effects varied with individuals' baselines. Some very old rats without baseline impairment showed less change. So these behavioral gains align with the di-hexasynaptic effects. Treated rats had significantly increased synaptic density and spine head size in the hippocampus, indicating stronger synaptic connectivity underlying
the cognitive improvement. And additionally, dihexa has shown neuroprotective effects. It improved the survival of neurons exposed to neurotoxic insults. Now we look at clinical studies. There are no published peer-reviewed clinical trials of dihecta itself in humans yet. However, a closely related compound known as ATH 1017, also known is phosgonymeton. I don't know who comes up with some of these names. which is essentially a pro-drug of dihexazaxiform is in clinical development for Alzheimer's disease.
And the mechanism is the same, which has through HGF agonism and preliminary reports show enhanced synaptic protein levels and some cognitive improvements in treated patients. The full efficacy results are pending. So the fact that the di-hexo derivative has advanced human trials underscores the translational potential of this approach. Then we look at the anecdotal report from biohackers. So some users describe Gradual with notable improvements in learning ability, memory and recall, and mental clarity when using dihex over weeks. And unlike stimulants, di-hexus effects are said to build up with time.
Again, that's why I would say it's more mild. With consistent change in the physical structure of the brain and common self-reported dosages are in 5 to 40 mg a day, often starting around 10 to 20 mg per day. I like the 10 milligram range. Most people will notice more pronounced nootropic effects at the 40 milligram dose. But when we combine that with the J147 and the Nupep, you can do a lower dose and get more bang for your buck. And many report minimal acute sensations, just steady cognitive gains, though a few note subtle side effects like nerve tingling or headache.
These accounts are subjective, but it does seem to have a brain building effect and it kind of accumulates over time, which is kind good when working using this product Biomind over Now, let's look at NuPept. So, NuPept is a small dipeptide-derived nootropic compound. It was developed actually in Russia at the Zakusov Institute of Pharmacology by T. A. Gudasheva as an analog of the racetam family of cognitive enhancers. So its design was actually based on the structure of paracetam, which is a very classic nootropic, if you're familiar with that world,
but is roughly a thousand times more potent than parasitam. Meaning the effective doses are much lower, so it has a short half-life, around five to ten minutes in rodents, yet produces longer lasting effects via the active metabolites once it is ingested. So one metabolite known as cycloprolaglycine or CPG is an endogenous neuropeptide that may mediate NUPEP's prolonged activity. So it's available as prescription cognitive enhancer in some countries like Russia and is sold as a supplement to others like the United States.
Nupep, let's look at the mechanisms. It enhances cholinergic cholinergic signaling. Man, I need some more neutropics. But it upregulates neurotrophic factors and modulates glutamatergic systems influencing the inhibitory neurotransmission. It has memory benefits, so it prevents memory loss induced by scopolamine, boosts acetylcholine function, and promotes neuronal growth and plasticity. It also protects neurons from glutamate toxicity, reduces oxidative damage, and inflammatory cytokines and brain injury models.
And it also has anxiolytic properties, so it has calming and anti-anxiety effects, enhances neurotrophins and neurotransmitter balance. So when we look at the evidence base, substantial body pre-clinical research, much of it from Russia, supports its pro-cognitive effects. In rota models of Alzheimer's, NUPEP, improved learning and memory. rats injected with beta amyloid to induce Alzheimer's like memory deficit showed improved performance and memory task after seven days of new pep, whether given preventively or as a treatment post injection. An olfactory model of Alzheimer three weeks of New PEP restored memory performance.
New pept has also blocked amnesia causing caused by inhibiting glutamate in DMA receptors and by colon and jerk cholinergic blockade indicating it counteracts diverse cognitive disruptions. Also at the cellular level, NUPEP increased BDNF and NGF levels in the hippocampus and cortex correlated with cognitive benefits. It has neuroprotective actions and vitro and increases the viability of neurons exposed to toxic levels of glutamate and reduces amyloid-induced cell death and tau phosphorylation can even inhibit aggregation of alpha-synuclein, which is implicated in Parkinson's disease and shows broad neuroprotective activity.
Humidate on NUPEP is still pretty limited. A small open-label clinical study in Russia followed 56 days of NUPEP, where they took 10 milligrams twice daily in patients with mild cognitive impairment. Results showed improvements in several cognitive domains and possibly mood, and the NUPEP group's MMSE score improved from 26 to 29 on average over two months. By comparison, a control group on paracetamol also improved but to a lesser degree, so a little bit stronger than that. And some subjective measures like reduced anxiety and better sleep were reported anecdotally in the study as well.
So although this trial lacked the placebo control, it indicates Nupep may benefit early cognitive decline. And then we would just look at, anecdotally speaking, it's widely used by nootropic enthusiasts and there's a lot of user reports on the internet. Many people like it around 10 to 30 milligrams per day. Now, again, in this case, we're using it at 10 milligrams because we are using with two other compounds. But in isolation, you probably want to do a little bit higher dose. And most people experience enhanced memory, recall, quicker thinking, and improved focus. They often describe a clear head-in-the-game feeling, increased motivation to mentally engage, sometimes mild psychostimulant-like effects,
like better concentration without the jitters. I'm all for that. Some biohackers cycle Nupec for neuroprotective benefits, citing its ability to increase BDNF and NGF as a way to support long-term brain health. And the consensus from anecdotal evidence is that it improves focus and mental clarity, Within days, so not over time like dihexa while memory memory benefits crew over long-term use so This aligns with the mechanistic findings that we discussed now Moving along to the cognitive benefits,
let's look at some of these. My clicker kept going off. So when we look memory and learning, J147, I'm gonna go through some different aspects of cognition. Let's looks at memory in learning first. All three of this compounds have shown pro-memory effects in research. In multiple studies, j148 improved learning and memory, especially in models of age-related decline. A hallmark result found that aged Alzheimer's mice tree with J 147 performed as well as young mice in memory tests, essentially rescuing memory function. It also enhanced spatial memory in normal age mice and object recognition memory and cognition impaired mice.
So this is from its ability to boost synaptic proteins and neurotrophic support of BDNF and NGF which strengthen the neural circuits underlying memory. The cool thing about this is it's pretty rapid whereas dihexanupec might be a little bit slower onset. So even in short-term treatment, reverse cognitive deficits and dementia model mice. And anecdotally, self-experimenters report better recall after a few weeks of J147 supplementation, though we don't have a lot of human data still. Dihexa is particularly noted for its effect on learning capacity and long-term memory formation.
So in rodent experiments, dihexatreated rats learned maze tasks significantly faster and retained memory longer than controls. And also mechanistically, by building new synaptic connections, DiHexa may increase the brain storage capacity, i.e. making our hard drive larger. Users who have tried DiHexa frequently report gradually improved memory recall and learning curve. which means they need fewer repetitions to memorize information and feel like their brain absorbs info like a sponge. Then we look at NUPEP in terms of memory. It's been shown to enhance memory encoding and retrieval in a variety of models.
In animal studies, it improved both short-term and long- term memory, And then also, Nupep usage correlated with improvements in memory-based test scores. Among Neutropic users, NeuPep is often praised for noticeably sharpening memory and people report remembering details like names, words, and facts more easily and experiencing more fluid recall of stored knowledge. So, it can be used as a study aid as well. And there's a lot of really cool benefits. Now, when we look at focus and attention, obviously a little bit different from memory, memory recalling, focus intention, we're focusing on what's in front of us.
So J147, the research is focused on memory and neuroprotection. Improved focus can be a downstream effect of better brain function and mood. So in age mice, J-146 reduced anxiety-like behavior, which is pretty cool. And anecdotally, people report a clear-headed feeling and increased mental energy when using J 147. When we look at dihexa, it's not a stimulant by improving synaptic connectivity. It may support the neural networks involved in attention. Users have described an ability to stay engaged in cognitive activities for longer when on dihexyl reporting improved mental clarity and sustained attention span.
So definitely more powerful in that sense than J147. Because di-hexa works in the background to optimize brain structure, the effect on focus is subtle and comes from having a healthier, more plastic brain rather than the acute neurochemical stimulation. When we look at these, one user noted a gentle enhancement in focus like my brain signal-to-noise ratio improved. Then we look at NewPEP. Among the three, NewPeP is one of the more commonly associated with improved focus and alertness in the nootropic world.
So although it was really aimed at memory, newPEPs cholinergic boost can lead to a better retention and faster information processing. In fact, NEWPEPS is often compared to mild psychostimulants without the peripheral stimulation, and users report it helps them stay focused for longer periods and provides a sense of mental drive. Some even find it to actually be slightly anxiolytic, which can give further improved focus by reducing distracting stress. So again, as with most nootropics, users' responses vary, but in this category, NUPEP is going to be better.
Now let's look at neurogenesis and synaptic plasticity, Which is basically just the growth of new neurons and how fast those can fire together. So J147 is strongly neurotrophic, so it elevates BDNF and NGF, it creates an environment conducive to neurogenesis. In aged mice, J 147 treatment increased the generation of new neurons in the hippocampus. It also upregulated proteins that are responsive to BdNf, which are involved in synaptic plasticity and memory formation. So basically J-148 helps the brain grow new brain cells and connections, essentially rejuvenating some aspects of brain structure,
Which is pretty cool. So dihexa has a standout effect on synaptogenesis, which is the formation of synoptic connections. So it has been described as enabling the brain to rewire itself by forming new synapses at an unparalleled rate. Lab data shows di hexa can induce spine and synapse formation, like we talked about, to levels comparable to or exceeding powerful growth factors like BDNF or HGF itself. By activating that CMAT pathway, di hexa triggers intracellular cascades that promote neurite outgrowth and spine maturation. This means neurons grow more branches and can and contact points with each other, which is foundational to learning and memory storage.
So while the effects on actual neurogenesis are less documented than it's on synaptogenesis, the increased trophic support could secondarily support neural precursor cells. It essentially acts like a brain fertilizer for synoptic growth, providing a structural basis for improved cognition. Now, we look at NUPAC. less pronounced, but still noteworthy. So, NUPEP has been shown to raise BDNF and NGF levels after about four weeks, which likely spurs neurogenesis in the hippocampus and BDENF is a key factor
for the survival and maturation of new neurons. Studies in rats have indeed observed increased immature neuron markers in a hippo campus with NUPEP, suggesting it aids in neuroogenic processes. And additionally, By reducing factors that impair neurogenesis, like excessive glutamate or inflammation, NewPEP creates a more favorable environment for brain cell growth. It also aids synaptic plasticity and memory tasks that rely on synoptic strength or improved, and neurochemical studies find that NewPeP can restore long-term potentiation. So there's even evidence that newpeps metabolite CPG directly exerts pro-neurogenic and neuroprotective effects.
Again, not as strong as some of the others, but still pretty cool. Now let's look at neuroprotection, basically guarding our brain cells. So J147 is a very potent neuro-protective agent. It was initially identified by phenotypic screening for compounds that protect neurons from multiple toxic insults associated with aging. J147 stood out when they were searching for this. So in vitro, it shields neurons against amyloid beta toxicity, collutamate, excitotoxicity, and oxidative stress. To animal models, J 147 reduced Alzheimer's-like pathology and treated Alzheimer mice had lower levels of soluble amylate beta and less synaptic loss than controls.
J147 also helps neurons repair and resist stress, and it not only improves cognitive performance, but also guards the neurons from damage again, which is why it's so powerful and being studied for the context of neurodegenerative diseases like Alzheimer's. So, dihexas' neuroprotective capability is closely tied to its regenerative property. By activating the HEFCMET pathway, an injury response system in the brain, it can help injured neurons recover and form new connections. Dioxa acts as a neural repair and protect agent.
It uniquely fixes what's broken by rebuilding synapses, and in doing so, protects cognitive function against progressive damage. And then lastly, NewPEP's tagline could well be nootropic and neuroprotective. So numerous preclinical studies support that NewPeP defends the brain from various insults. In a cellular model of Alzheimer's, NewPEP significantly reduced neuronal death. For example, after traumatic brain stress or ischemia, it decreased the levels of pro-apoptotic signals and reduced inflammatory cytokines.
And also, NewPeP treated animals showed less cognitive deficit and neurona loss than untreated ones. So NewpeP also prevented glutamate-induced neurono damage, which preserves cells' viability. and you can get an immediate cognitive boost from it but as a long-term brain health supplement it aims to leverage its antioxidant and neuroprotective effects. So all pretty effective in that sense. Now let's look at some of the side effects so J147 as we've seen so far has shown excellent safety profile on animal studies with no significant toxic
effects at effective doses. I think this is important to examine in light of any time we're modulating things in the brain because obviously we don't want to go in a bad direction that you could with a lot of brain-altering compounds. So, in theory, because J147 modulates mitochondrial function, extremely high doses could affect cellular energy metabolism, but no overt signs of mitochondria toxicity have been seen in vivo. One interesting fact is that J 147 can reduce melanin synthesis, Those are only noted in one study at higher concentrations and its relevance to human
use is unclear. I haven't heard anything about that but when I was researching that was the one thing that I could find. So obviously long-term safety in humans is unknown and as a multi-target agent there could be unanticipated effects with chronic use. so again use with caution. i'm going to talk a little bit about how to cycle these at the end of the presentation. There are no drug interactions that have been documented for J-147 in a mouse study. Co-administration with Donna Pizzil was what I said was actually beneficial and did not cause harm.
However, just be careful when you are doing it and obviously with the dosing go low and slow. When we look at dihexa, it's shown minimal acute side effects in animal studies and rodents tolerated well at cognitive-enhancing doses. Biohackers often report that dihxa is subjectively well-tolerated. I've very rarely heard anyone say anything bad about it other than they just don't get any sort of effects from it. But some users have mentioned mild headaches or irritability on days they take higher doses, Now, when we look at risk, the main concern lies in its mechanism.
So because it activates this HDFC met pathway, involved in cell growth and regeneration, there's a theoretical, not actual, but theoretical risk that long-term di-hex use could promote tumor growth. However, at the doses we're talking about, I would just be very, very... How would I say? I wouldn't be so worried about that. If we are chronically stimulating this pathway through dihexa or other means, There's possibility, even though it's unproven, of increasing, cancer risk or accelerating existing tumors. It's important to note that no tumors or cancerous changes were reported in the animal studies of dihexa.
So again, just again it's a theoretical concern. I've made videos talking about the theoretical versus the actual risk, but by stimulating that pathway, that is something that I will just put out there that you'd be aware of. There's very little data, some sources recommend not stacking diheexa with other neutropics or psychoactive substances due to its unique mechanism. Now in this case, I really like it with J147 and NuPept, but again, it's not something that you want to take a bunch of other nootropic compounds with.
And so again, more research will need to be done, but I would say it's relatively safe for what we're talking about. New PEP in Russia, many studies have shown it to extremely well tolerated at recommended doses. So in that 56-day human study that was talking outside effects for mild and infrequent, five of the 31 reported sleep disturbances, three reported irritability, and seven saw mild increase in blood pressure. But overall, it was considered safe. and with fewer complaints than the other control drug, which is paracetam. So again, the typical dose was 10 to 30 milligrams a day, and that really caused issues.
Now, potential side effects, we could have slight anxiety or restlessness occur in very sensitive individuals, where others find it actually reduces anxiety. Again, this is where neutropics, two people could take it and have completely opposite effects. But importantly, new PEP does not appear to have significant toxicity. And when we look at long-term use, there is an extensive data beyond a few months, given a short staff life. It doesn't accumulate, which is reassuring for long term safety. So, New Pept is often stacked with other nootropics and generally plays well. For instance, when pairing with a choline donor like AlphaGPC, it can enhance memory and even prevent headaches.
There are also no known dangerous interactions, however, because NUPEP has mild MAO inhibiting effects in the cortex from some studies, one should be cautious combining it with other MAOs inhibitors or stimulants to avoid blood pressure spikes or agitation. So overall, Nupep has a very low risk profile. Now, let's look at the synergistic effects we're going to get from stacking these because this is where it comes in. All of these could potentiate and actually enhance the other effects, which one allows us to stay at a lower dose, but then also two allows to maximize
the effects with minimizing side effects. So when we look at the complementary pathways, each compound acts on a different primary pathway. J-147 on the mitochondrial AMPK and broad neurotrophic signaling, dihexa on this HGF-CMET synaptogenesis pathway, and then nupept on cholinergic and glutamate glutamatergic modulation plus the BDNF-NGF upregulation. So, when we use all of them together, there is a synergy by covering multiple bases. For example, J147 and dihexa both promote neuroplasticity, but via distinct growth factors.
In theory, using them might yield an additive or synergistic effect on neuron growth. J-147 could enhance general neuronal health and energy availability, making neurons more responsive to the dihexas synaptogenic push. Meanwhile, di-hexes synapses might benefit from J 147s in maintaining those connections through mitochondrial protection and anti-inflammatory action. And then we bring in NUPAC, it adds another layer by enhancing neurotransmitter signaling through acetylcholine. It could help functional integration of the new synapses.
So in short, J147, dihexite and Nupac provides a broad spectrum cognitive enhancement through structural growth, trophic and metabolic support, and neurotransmitters optimization. When we combine these together, they all kind of enhance each other. J147 and NuPep both provide neurochemical support that could amplify dihexas structural remodeling of the brain and J 147 plus Nupep cover both long-term and short- term enhancement. So it's kind of triple punch that kind gives us a short term boost plus a long term brain.
health and regrowth situation, which is really cool. And so when we consider these, obviously use at your own discretion, but sometimes less is more. Again, that's why we can stay on lower dose of these and really get the effect. I did have a comparison chart here. So we look at J147. That's more of the mitochondrial ATP side of things. It reverses memory loss. it has an angiolytic effect and is neuroprotective. and prevents oxidative stress, helping with things like TBI. Dosage, if you're using it in isolation, 5 to 30 milligrams.
Side effects, very well tolerated, and there is strong preclinical evidence that leads us to believe it's going to have lots of good effects for people. Dihexa, again, it is an HEF slash CMET pathway. Activator binds to HEf, promotes CMet signaling, induces synaptogenesis and dendrite growth. dramatically improves spatial memory and impaired rats. It also drives new spine and synapse formation and supports neuron survival in Parkinson's disease and stroke models. Again, dosage wise, 10 to 40 milligrams.
very minimal side effects, you might get a headache in higher dose use situation. And then there's really good data around it that we have so far in animal studies. In the new PEP, very multifaceted, it modulates glutamate and ACH receptors. There's neurotrophic factors in the hippocampus and then antioxidant anti-inflammatory action through the lowering of excitotoxicity in IL-6. It enhances memory consolidation and recall, it improves attention and mental clarity, and then there is a neuroprotective aspect. And again, 10 to 30 milligrams in isolation is usually the dose there.
Now, what is my protocol? So you might be skipping ahead to this part of the presentation, but What I recommend doing is only using this product by a mind two to three times a week. So this is going to kind of give us a pulse method to high-impact neuro enhancers rather than flooding the brain daily. One, it reduces the risk of overstimulation. All three compounds, especially dihexanupep, modulate powerful growth in neurotransmitter pathways like BDNF, NGF HGFCMAT,
and glutamate in acetylcholine signaling. If we were to use this every single day, eventually what could happen is we could desensitize receptors. We could drive too much synaptic growth. we can raise neurotrophin levels beyond the optimal. BNF is a Goldilocks molecule. Too little is bad, but too may disrupt emotional stability and cause excitability or anxiety. And so by using this just two to three times a week, we allow for synactive changes to integrate and consolidate the nervous system to recalibrate and a safety buffer from potential unknown long-term risk, which I think is very low, but still we're not driving neurogenesis too hard.
And I, think if you were to take this every day, again, in a use case, you might need it, But over the long term, I would not use this ever single day. So again as someone that sells this product, that doesn't behoove me to say, don't use it every single, day but I'm telling you, You really don' want to use more than three times per week and that's personally what I do. I only use two to three time per weeks. So, just like with training our muscles, pulsing cognitive stimulation allows time for repair and regrowth. So you get the stimulus, which is the synaptogenesis, the neurogenesis and mitochondrial enhancement, followed by a recovery phase where the brain adapts and up-regulates itself. Now, what do I do on those non-days?
Well, thanks to Alex Kickle, who kind of came up with this protocol, I use AlphaGPC. Alpha GPC is a bioavailable colon donor. And precursor to acetylcholine, which is a key neurotransmitter for memory, attention, and plasticity. So here is why this is really good to use 600 milligrams of alpha GPC on non-use days. We can sustain the cognitive performance without pushing plasticities pathways too hard. On non use days of BioMind, you're not heavily stimulating BDNF, HDF mTOR and neurogenesis, but we're still supporting neurotransmitters production through acytocholene.
We're still aiding mental clarity and working memory, and we're reducing choline depletion symptoms like brain fog and fatigue that can occur after using NUPEP. We also protect and potentiate synaptic changes, so cholene is required to maintain the structure of cell membranes and synapses. And after dihexan J147 create new synapse, alpha GPC may help nourish and maintain them. So it also supports REM sleep and memory consolidation, enhancing integration of neural growth from prior days. So we also, more importantly, avoid downregulation of choline receptors.
Overusing cholinergic compounds like nupept or racetans without replenishing cholene can lead to receptor burnout, and AlphaGPC helps buffer this and ensure your brain is topped off. That is it for the slides. And that is my deep dive into bio mines ingredients, which are J 147 dihexa and new pepsi. Hopefully it was helpful to you. I know it's a little bit long winded, but I really want you to have a good context and framework for what these three different compounds are doing. And we're using two capsules, 10 milligrams, five milligrams of each per capsule.
If you use two, it'd be 10mg. first thing in the morning, two to three days per week, and then on the non-U stage, use 600 milligrams of Alpha GPC. And I've been doing that for a few weeks now and it's worked amazing. I really, really love BioMind. Again, I'm not a nootropic head by any means. To me, always kind of steer away from nootypics because they make me anxious and make my brain move too fast. So I don't really like that effect, but I had a really good response to Bio Mind. Definitely has improved cognition for me. It seems to have improved my ability to read and learn, which was already pretty good, but I will say it definitely has an improved thing there.
I think for me too, the J147 is really cool. In light of anyone that's experienced head trauma or anyone who is experiencing some sort of neurodegeneration as you get into old age, I that could be a really powerful game changer for brain health there. So the feedback from the community so far seems to be amazing. People really, really love this. Again, it's just one of those things. You want to make sure you're not using every day and you are making sure your replenishing acetylcholine through the use of alpha GPC on non days. So hopefully that was helpful to you guys.
