ACE-031 Myostatin Inhibitor for Faster Gains Worth the Hype
I recently got my hands on ACE-031, a peptide that's been quietly floating around bodybuilding circles for years. Someone told me it's going to blow up in popularity over the next couple of years, so I figured I'd test it on myself before that happens. Here's everything I learned, plus my honest take on whether it's worth your money.
What ACE-031 Actually Does
ACE-031 is a myostatin inhibitor. Originally developed to treat muscular dystrophy, it's now getting attention in biohacking circles for its muscle-building properties.
Here's the basic idea. Your body produces myostatin to keep itself in homeostasis. The body doesn't actually want to build muscle. More muscle means more energy expenditure, more calories burned, more work for the body to do. So myostatin acts like a governor, limiting how much muscle you can put on.
ACE-031 binds to the ACT-R2B receptors and blocks myostatin from doing its job. In theory, this takes the governor off and lets you build more muscle than your body would normally allow.
It also targets GDF-11 (involved in cell aging) and other muscle-limiting proteins. The peptide isn't actually building muscle directly. It's just disrupting the process your body uses to keep muscle growth in check.
The Potential Benefits
- Increased muscle mass
- Greater strength
- Improved metabolism (more muscle means better insulin sensitivity and fat burning)
- Better bone density (early animal studies)
- Faster recovery from training
The recovery piece is the one I actually noticed in my own experiment. More on that in a minute.
Side Effects to Know About
Most peptide side effects sound similar, but ACE-031 has a few unique ones worth knowing.
At higher doses in clinical trials, some users reported nosebleeds and gum bleeding linked to effects on blood vessel growth. Blood pressure can increase with extended use. Red blood cell count can also go up, which is a double-edged sword. If you're training hard, that extra oxygen delivery is great. If you're not exercising, you could end up with thicker blood and cardiovascular issues.
Muscle cramping is also possible as your tissue adapts. I deal with cramping anyway because I sweat a lot, so I can't say for sure if that was from the peptide.
I personally didn't experience nosebleeds, blood pressure issues, or rashes. The injection itself didn't sting.
What the Research Actually Shows
There are really only a few human trials worth mentioning.
Phase 1 trial (2010-2011): Healthy volunteers received single doses ranging from 0.02 mg/kg up to 3 mg/kg. They saw increased lean muscle mass and strength, with mild side effects like nosebleeds at higher doses.
Phase 2 trial in boys with Duchenne muscular dystrophy: They saw initial muscle improvements but the trial was halted due to safety concerns. Doses were 50 mg/kg every 4 weeks or 1 mg/kg every 2 weeks.
Post-menopausal women study: Researchers found significant increases in lean body mass and thigh muscle volume, plus improvements in bone and fat metabolism markers. Dose was 1 mg/kg as a single injection.
To put that in perspective, I weigh 220 pounds (100 kg). At 1 mg/kg, I'd need 100 mg in a single shot. ACE-031 typically comes in 1 mg vials at around $160 each. That's $16,000 for one injection. Completely cost prohibitive.
How I Dosed It
I used a 1 mg vial split into 250 mcg, twice per week. One vial lasted me two weeks. I ran it for a full month.
Most people cycle ACE-031 for six to eight weeks with breaks in between. The half-life is around 10 to 15 days, so you can dose less frequently. I prefer to spread doses out to keep blood levels stable rather than slamming a big shot once a week.
Injection can be subcutaneous or intramuscular. I did sub-q. If you wanted to target specific muscle groups, intramuscular into those areas might give you some satellite cell activation, but I didn't test that.
My Actual Experience
It definitely did something.
Within the first week, as it built up in my system, I felt bigger at the gym. Recovery was noticeably faster and I was much less sore between sessions. I'd estimate I was around 5 to 10% stronger than I normally would have been.
Did I put on noticeable muscle mass? Not really. I didn't do a bod pod scan, but visually and on the scale, no big change. I kept my testosterone, growth hormone, and other peptides at maintenance levels during this run so I could isolate the ACE-031 effect.
My Take
For the price and the dosing required to see real results, I don't think ACE-031 is worth it right now.
If you wanted to put on muscle affordably, increasing testosterone and growth hormone gets you much better results, much faster, with way more research behind the safety profile. People run mega doses of test and growth hormone all the time without major issues. ACE-031 just doesn't have that track record yet.
I'm also generally not a fan of inhibiting the body's natural processes when I can add things in instead. I'd rather supplement a hormone than disrupt a regulatory protein. That's a personal preference, not a hard rule.
If you don't want to use testosterone or growth hormone, ACE-031 is a peptide alternative worth experimenting with. But for me, the cost-to-benefit ratio just isn't there. It did something, but not enough to justify what you'd need to spend to really see big results. Maybe in a few years when the price drops and we have more data, this changes. For now, it's a meh from me.

Today's video is going to be about a peptide called ACE-031.I'll probably just call it H-31 because that rolls off the tongue a little bit better. But this a peptide I recently experimented with. It's not as popular and the reason is because it's much more expensive than a lot of other peptides. It's obviously much less known than some of the other peptides, but there's a few research chemical companies that do sell it.
So I actually ordered this recently because someone had mentioned to me that this is going to be up and coming in the next couple of years as it gains in popularity and probably comes down in price a little bit. I figured, Hey, I'll give it a shot. so I ordered some myself and tested on my lab rat, so to speak. uh, to see how it would do. And I ran that and wanted to make a video about my experience with it. So H 31 is a peptide that basically works to inhibit myostatin, which I'm going to go into on this video, but in the process of doing so,
The thought around inhibiting myosin is that it will help increase your muscle mass and in doing so also help you burn fat to basically just have a better for the sick. So I wanted to put this to the test. Um, hopefully this video will kind of be like a roadmap and guide map for your own research purposes. Because prior to this, there really wasn't anything I could find that specifically gave parameters around a use case and dosing. So I kind of just went off of studies that I was able to procure myself, which I'll talk a little bit about in this video to figure out the dosage and
use cases and everything like that. Also, if you stick around the end, I give my own personal experience. I'm going to go through everything you should know about it, but then also to just what I experienced. if I would recommend doing it again and what my plans are for in the future. I am Hunter Williams, and this is the comprehensive guide to ACE 31. And we're going to discuss myostatin inhibition myths versus reality, because this something that will probably come to light much more in the future. It's been in bodybuilding circles for a while. So I just wanted to do a breakdown of that to give you guys insight from my unique perspective. What is ACE-31? It basically is a therapeutic peptide.
This recently gained a lot of attention for its potent muscle building properties. And like I mentioned, the way it works is by reducing myostatin activity, which apparently is supposed to lead to significant gains in muscle mass and strength. For most people, this would be kind of targeted towards your bodybuilders, athletes. And also it was originally studied for people with muscle wasting disease. So they're going to look to myostatin inhibitors like ACE31 is another pathway that they can potentially tap into to build muscle. And the benefit is that it enhances muscle growth beyond our natural limits and improves performance and obviously leads to quicker recovery.
So what does this idea of like myostatin inhibition mean? So ACE31 is a myostatin inhibitor. It was developed to increase muscle mass originally for people with muscular dystrophy. But basically it blocks the body's natural limitations on muscle growth. So it disrupts myostatin's interaction with its receptor, which allows muscles to grow more freely. Basically, to give you kind of like a framework, the buddy produces myostatin to keep it in, I guess, homeostasis, you could say.
The body does not want to build muscle. It is not natural for the party to put on the muscle because in order to do so, your going to have to expend a lot more energy. So the body naturally doesn't want as much muscle. It will respond to stimuli. But in terms of keeping the in a state of homeostasis, the more muscle you have, The more calories you're going to burn and the energy you are going expend. And when we have more Muscle, body has to work way harder. So as you try to put on muscle, Body is going really pull back. Anyone that knows and has tried to Put on appreciable amounts of muscle in their life, you know how hard that can be.
Regardless of the drugs or supplements you taking. Basically the idea is that myostatin blocks this process to keep the body in homeostasis. So when we do so, the thought is, that we're kind of taking the governor off the to build more muscle. Originally developed for treating conditions like muscular dystrophy, but in doing so it's gained popularity in the biohacking communities for its ability to enhance muscle development and strength. So let's look at the mechanisms of how ACE-31 works, especially as it relates to myostatin.
And so it binds to what are known as the ACT-R2B receptors, preventing myostatin and other muscle growth inhibiting proteins from activating these receptors. Myostatins normally signals the body to slow down or stop muscle gross growth, but ACE 31 disrupts the communication between the receptors allowing for continual muscle growth and increased muscle mass. So when we look at these proteins, obviously myostatin is the primary target, which inhibits muscle. We've also got GDF 11, a protein involved in cell aging and muscle function, but also limits muscle which are a group of proteins that regulate processes
like inflammation metabolism, which can also affect muscle tissue. So by inhibiting all of these proteins, ACE31 creates an environment in the body that is more conducive to muscle growth and repair. The peptide itself is not breeding muscle tissue or anything like that. Basically it's working to suppress these proteins to disrupt the process of the body naturally goes through to limit muscle growth in order to keep you in a state of homeostasis and energy conservation. So, uh, you know, as opposed to anabolic or growth hormone peptides or something like.
This is actually disrupting a process. The body, naturally. has in order to grow muscle. So that's kind of the way it works. Now, what are the benefits of that? Well, first, we're going to have increased muscle mass. In theory, if we use this, it's going stimulate muscle growth, which for conditions like muscle wasting diseases is an attractive option, but it also is good for performance enhancement. In the process of doing that, it's also going to increase strength. So increased muscle mass also leads to greater physical strength, benefiting not just athletes, but also the muscle loss due to injury or illness.
It's going reduce fat, so enhanced muscle growth basically increases your metabolism. so it doesn't directly burn fat. But when we have more muscle on the body, that increases our metabolism, thereby making us more insulin-sensitive and better able to burn fat, which is going to obviously make us leaner. And also, this is especially true for aging individuals, can improve bone density. So early studies in animals suggest that H31 may also improve bones density, making it potentially therapeutic for things like osteoporosis.
I'm going get to that in a second. And obviously, too, it helps speed up muscle recovery following injury or intense exercise, which is ideal for athletes. And I will say, to spoil a little bit of my own personal review, I did notice this relative to some other things. So what are some of the side effects? So obviously with a peptide, Most of the side effects are always going to sound the same. There are a few other ones in this that could potentially come up, not that I experienced.
So we do have injection site reactions. I personally, this peptide did not sting. It did cause me to have a rash or hives or anything at the injection sites like some other peptides, but I always throw that in as a disclaimer. When you're dealing with peptids, the possibility exists. This one, this, I never felt anything like this but in higher dosages of this and studies that they use on children. Some individuals reported nosebleeds, which can be linked to a 31 effects on blood vessel growth. There is the potential for increased blood pressure.
So the peptides influence on vascular tissue growth can cause elevated blood and some users, especially if used over extended periods. I think in terms of looking at blood pressures, very similar to antibiotics, how when we use higher doses of antibiotics that can increase blood And when you're doing the same thing, the potential there exists. Uh, it can increase red blood cell count, which can be a catch 22 in one way. It could be good, but then if you not exercising and doing cardio, in theory, It can lead to your blood being thicker, um,
and cause cardiovascular complications. So, Again, in doing so, it's going to improve oxygen delivered to muscles, which for athletes and active people is good. But if you're not active, this could potentially be a downside because you are increasing red blood cell count, Which could lead to a thicker viscosity of the blood if that oxygen isn't used properly in the form of exercise. And I did notice a little bit of this. So muscle cramps, so rapid muscle growth may cause discomfort or cramping as muscle tissue adapts to the peptides effects.
and kind of raising our metabolism and the byproducts of doing all this, cramping is a thing. I struggle a lot with crampping just because I'm someone that sweats a lots. So, you know, that potential is there, but I didn't notice any nosebleeds. And as I want to talk about, the nose bleeds came in at very high doses. Let's look at some of the research around this. These were the only human trials I could find, there was a phase one clinical trial on healthy volunteers from 2010 to 2011. This found that ACE-31 increased
lean muscle mass and strength without severe side effects. So again, the minor issues such as nosebleeds, gum bleeding, and blood vessel dilation were reported and those were the safety concerns. But the trial was successful in enhancing muscle loss, but mild side-effects led to further safety investigations. So participants in this study received 0.02 milligrams per kilogram of body weight, 0,1 milligram per kilograms of a bodyweight, and 0.,3 milligram for kilogram bodyweights. And they also did another batch of test subjects that did 1 milligram, 3 milligram.
resulting in notable increases in muscle mass. So they also used a single injection. I think this one, they just injected one time because it has a very long half life. And I they monitored for several weeks, like four to six weeks after. So to give you an idea of like what that would look like, I'm 220 pounds, so I am 100 kilograms. So for me, i would just take those milligram amounts and then multiply them by 100. For instance, if I did 0.02 milligrams per kilogram,
basically that um, milligrams total. So I would do one injection of two milligrams. Um, if it was 0.1, I will do 10 milligrams that was point three. I'll do 30 milligrams and then if I did one milligram per kilogram, that'd be a hundred milligrams, which as you'll see, it's really cost prohibited because it is typically sold in a one-milligram bio. If that's like 160 bucks, you'd have to have a 100 of those vials to do. It just would be cost prohibitive and not useful.
Unless you, I guess just money was like not an issue whatsoever and you wanted to spend $1,600 per month on a peptide that may or may not work. So the next part of this was phase two clinical trial in boys with Duchenne muscular dystrophy. Um, so the, they did this original trial and they moved into looking at it for people with muscle muscle wasting. Uh, initial improvements in muscle mass were observed in these boys, but the study was halted due to safety concerns. Again, the nosebleeds and blood, blood vessel dilation. Um, and they suspended the trial, but basically the dosage here, they received, uh, 50 milligrams per kilogram every four weeks and one milligram per milligram
every two weeks. So just to give you again, an idea, if I'm doing those dosages, I basically doing either 50 kilograms or a hundred kilograms, excuse me, a 100 milligrams, which again would be like 50 vials or 100 viles all at one time. Uh, so obviously that's going to have some side effects. And lastly, there was a more recent study, I think this one was 2016. I forget to put the date on here when I was compiling everything, but the objective in this was to study the effects in post-menopausal women.
So they found that significant increases in lean body mass and thigh muscle volume were observed along with unexpected improvements in bone and fat metabolism biomarkers. It actually worked really well in his case. Age 31 demonstrated potential to maintain muscle mass. and improve bone and fat metabolism, but further research was continued due to safety concerns. And again, this was at one milligram per kilogram. So again that's a very high dose, 100 milligrams per kilograms. Um, and this one, they just did one injection and they monitored everything after four weeks.
Um, as you can see, uh, the dosage is kind of like all over the place here. And I think as we always say, The difference between the pill and the poison is the dose. So that leads me to the doses. Now, when I use this, I got a one milligram vial and a 250 micrograms, two times per week. That one vile lasted me for two weeks. Again, it's $160 typically retail and it just becomes cost prohibitive, for the effects I've got at that dose Typically,
people are going to cycle this for like six to eight weeks with breaks in between to assess progress and side effects. I did give this one a month, so I used it for a months. From what we can gather, it's around 10 to 15 days, meaning that remains active in the body for an extended period of time after administration. This is why they were able to give the post-menopausal women one shot and it lasted and they saw improvements over those weeks.
So this allows for less frequent dosing typically once a week. However, if you know anything about me, you'll know that I typically, in order to stabilize levels of everything, I tend to go lower and more frequently instead of injecting things once per week or even once every two weeks or four weeks like they were doing in the studies. However because of long half-life, the effects may continue to linger in body for several weeks after stopping the peptide. So just pay attention to your timing. And then injection, as it usually goes, people will do this subcutaneously or intramuscularly.
I just did this up cutaneously. There probably may be some sort of like satellite cell muscle activation if you inject it directly into the muscle. or muscle groups that you would like to grow. I didn't do that in this case, but just make sure you're obviously researching properly when it comes to injection. And lastly, I just want to give my experience, which is everyone wants to know. Yes, this is a slide with not a lot on it. It definitely did something.
Within the first probably week or so, as it started to build up within my system, I definitely noticed it was doing something. I felt bigger at the gym. Um, I'd definitely recovered quicker. So I was much less sore. And I think even for, for that use case, it could be beneficial. The difference here is probably the dosing because the dose things all over the place. Again, if it comes in a one milligram vial, you could probably do like one milligrams per week and see more pronounced benefit.
Obviously that's still under, a lot of the dosing that we saw in terms of side effects in the studies. But I think when we talk about picking our battles and choosing our battle when it comes to building muscle and burning fat, whatever, I just think there's a better ways to spend your money and better pathways to hit. I also didn't mention this, but I, again, I'm not saying this is wrong, but I am typically one to not be the biggest fan of inhibiting the body's natural processes.
And I know we're biohacking, so you can get into the weeds, and I notice people say, well, there's a bunch of stuff you do that inhibit natural process, like Metformin or things like that. Yes, that would be correct, But I think when we look at it, tendency to think that it's probably a little bit better or more reasonable to add things in, like add in hormones, then disrupt ones. And I know my, my sense on a hormone, it's a protein. But I think when we look at that, this is kind of like, eh, I don't know if it is necessarily the best thing to do.
I there's other ones out there. There's things like YK11 is one that people talk about that does real well. That's cool. In my experience, for the dosage, of what it did. I just don't know that I got that much out of it. So I'd say for me, it didn't really like put on any muscle mass. Now I didn' do a bod pod to really measure out. And obviously I do other things like testosterone growth hormone and all that stuff, but I kept those relatively stable during those times and was just like, okay, let me see what this does. Meaning that with the other stuff I wasn't tweaking or doing anything different than what I would normally do, just kind of in the maintenance phase.
In my experience, I don't know that it's necessarily worth the money at the doses that you would get and have that would be cost prohibitive, meaning that if I really wanted to put on muscle, for an affordable rate, could increase testosterone and growth hormone and get much better results much faster without any concerns about side effects than I could with ACE31. Now, someone would look and be like, well, maybe I don't want to use testosterone growth hormone. Okay, that's fair. You could try H31 because it is a peptide, but I think when we look at safety and efficacy, there's way more research around people using mega doses of
testosterone and mega dose of growth hormones. without really any negative side effect as compared to this where it's just not there. So maybe something into the future, it becomes something that is much better. Um, so I don't want to say this was worthless cause it definitely did something. I think it like helped with growth slightly in terms of like helping me recover faster and better so that my lifts were better and I would say I was probably like five to 10% stronger. than I normally would have been otherwise. So I think it was good overall. But it just, again, when it comes down to efficacy of cost versus anything else that I could potentially use in my toolkit,
I just don't notice there. I'm not saying don' try it. Definitely experiment with and research on it and see how you like it, but I would just say relative to the price and everything, it's just kind of, meh, that's how I felt. That is it for the slides.
