SS 31 - The Most Underrated Peptide A Complete Guide
SS-31 might be the most slept-on peptide in the biohacking world right now. I've talked about it before, but I wanted to bring this guide back because it deserves way more attention than it gets. Here's everything you need to know about why it works, what it does, and how to dose it.
What SS-31 Actually Is
SS-31 is also known as Elamipretide. It's a synthetic tetrapeptide, which just means it's made up of four amino acid residues. The chain includes D-arginine, dimethyltyrosine, lysine, and phenylalanine.
That specific sequence matters. It's what gives SS-31 its ability to target the mitochondria directly, which is where the magic happens.
How It Works
SS-31 binds to the inner mitochondrial membrane. That sounds technical, but the practical effect is huge.
First, it fights oxidative stress. If you've been around my channel for a while, you know oxidative stress is one of the main drivers of aging. Our environment is loaded with pollution and toxins that create free radicals our bodies can't keep up with. By reducing oxidative stress, SS-31 gives cells a fighting chance to maintain function and stay alive longer.
Second, it boosts mitochondrial function directly. More efficient energy production means healthier cells, which means a healthier body.
Here's where SS-31 stands apart from something like MOTS-c. MOTS-c helps with energy production at the cellular level. SS-31 actually goes inside the mitochondria and works on their function. If you've gotten benefits from MOTS-c, you'll likely get even more from SS-31.
The Benefits
Ischemia-reperfusion injury. When blood flow stops and then returns suddenly, like during a heart attack or stroke, the cellular damage can be massive. Early studies suggest SS-31 may significantly reduce that damage and speed up recovery.
Neurodegenerative disease. Most of us know someone dealing with Alzheimer's or dementia. I do too. The brain runs on mitochondrial function, and when those mitochondria fail, brain cells start breaking down. That's how you get brain fog, memory loss, and eventually full cognitive decline. Parkinson's and Alzheimer's are both linked to mitochondrial dysfunction, and SS-31 has shown promise in slowing progression and easing symptoms.
Age-related disease. Aging is basically the slow accumulation of oxidative stress and declining mitochondrial efficiency. Conditions like macular degeneration and sarcopenia (muscle wasting) both benefit from SS-31's mitochondrial targeting.
A lot of people have also reported chronic conditions like fibromyalgia, MS, and Hashimoto's improving on SS-31 because of how it restores mitochondrial function.
Side Effects
Side effects are rare, and the ones that do show up are pretty standard for any peptide.
Some people get mild GI upset or nausea right after injection, usually fading within 5 to 10 minutes. A few report mild headaches, though it's unclear if SS-31 is the actual cause. You might also see a small rash at the injection site, which usually has more to do with injecting than the peptide itself.
Dosing
Information on SS-31 dosing is hard to find. I had to dig around with some insider sources to put this together.
The standard dose is 10 mg per day, injected subcutaneously into the belly fat with an insulin needle.
I'd take it fasted first thing in the morning and hold off on food for 90 minutes to 2 hours after. That seems to give you the best cognitive and neuro benefits.
For the cycle, run it 8 weeks on, 8 weeks off. Mitochondrial peptides have lasting effects because of how they upregulate mitochondrial health, so you don't need to stay on them year-round. Three cycles a year is plenty for most people. That keeps your receptors fresh, avoids antibody buildup, and keeps the cost reasonable.
For acute conditions, some people have gone up to 50 mg per day. I've heard of folks with kidney disease running that protocol and seeing their condition essentially disappear. It's powerful stuff.

My Take
SS-31 is the most underrated peptide in the underground biohacking space right now. The mitochondrial targeting is something most other peptides just don't do, and the downstream benefits cover everything from heart health to cognition to chronic disease. It's a little more expensive than other options, but the results are worth it. I'm excited to run it myself and see what happens.

Basically, just to give you an overview, SS 31 is also known as known As a Lampertide, and it's a synthetic tetra peptide. So tetrapeptide basically just means it because it commits consist of four immediate amino acid residue. Arge, DMT, lysine, phenylalanine NH2. Basically just that means that there's four different residues from it. D form of the amino acid arginine DMD, which is not dimethyltryptamine, but dimethyltyrosine lysines and then phenyloalanines.
So it's just a combination of those amino acids in specific form. And that's what most peptides, they're just chains of amino assets that get a desired result in the body. So the specific sequence and modifications are thought to be critical for its mitochondrial targeting and capabilities for pharmacological action. So now that the technical stuff's out of the way, what are the mechanisms of SS 31? So, the primary role is centered on the inner mitochondria membrane and its interaction with the membrane has several implications. The first one being combating oxidative stress. If you've been watching my channel for any amount of time now, you know that combing oxidant stress is one of most important things we can do.
Oxidative stress is the main reason that we age. So a lot of the aging processes that take place in the body come from oxidative of stress. One of significant challenges we face, especially in our environment today, which has a lots of oxidate of pollution is damaging free radicals and they basically overwhelm our body and we can't defend against them. Oxydative Stress has been implicated in everything from premature aging to a bunch of diseases, usually the disease is associated with aging. So here's where SS 31 is so great. By reducing oxidative stress, it offers cells a fighting chance against various detrimental effects, preserving not just their function,
but their very integrity. Basically, more or less, we're extending the life of cells, which will extend our lifespan and more importantly, our health span. It also enhances mitochondrial functionality. Beyond Defending against the threats of oxidative stress. It also holds promise and actively boosting mitochondrial health. So when we enhance the mitochondria function, we're basically enhancing the ability to up-regulate energy within the cell. We're going to have more efficient energy production, healthier cells, and by extension, a healthier body.
So some of the benefits that specifically SS-31 is going to help with, first one's going be ischemia reperfusion injury. So in events like heart attacks or strokes, the sudden stop and subsequent return of blood flow can cause excessive or extensive cellular damage. Preliminary studies suggest SS 31 might significantly mitigate this damage, making recovery faster and reducing longer-term complications from some these traumatic events, especially to the heart. Also, we have neuro generative disease. So obviously, most people now have someone in their family that has experienced neuro degenerative seas like Alzheimer's or dementia,
myself included. Basically the brain with all the neurons relies heavily on efficient mitochondrial function. So one of the reasons people get neurodegenerative disease is because of impaired mitochondria function, the body cannot get enough energy into the cells of brain and these cells start to break down and we lose function and get brain fog and eventually lose our memory and cognition. So these diseases like this, like Parkinson's and Alzheimer's have been linked to mitochondrial dysfunction, obviously. So SS 31, because of the way that it helps with mitochondria function has been shown to help with these conditions and provide therapeutic benefits,
potentially slowing disease progression and alleviating symptoms. Which brings to the next benefit, which is age related diseases. Basically aging results from oxidative stress And this naturally brings a decline in mitochondrial efficiency. So conditions like macular degeneration, which is basically just the degradation of your eyesight or general muscular weakening like sarcopenia, just basically the breakdown of muscle and the loss of muscles in the body, eventually will cause eventually death, but a lot of diseases beforehand benefit
from the ability of SS31 mitochondria targeting effects. And where, you know, SS 31 stands out differently as opposed to a mitochondrial peptide like MOTC is that it actually goes into the mitochondria and works on their function as supposed to just assisting with production of energy within the cell, which is really cool. So if you get good benefits from SS 30 or Motsi, You're definitely going to get a good benefit from S S 31. So we're some of the side effects.
These are pretty much par for the course of side-effects from any peptide. They're very rare, but if you do have a side effect from a peptid, they're usually one of these three things. So you're going to have maybe some sort of digestive upset. You might just experience a little nausea or GI disturbances. If anyone has used peptides, you know, that's just par of course is something you'll deal with. And a lot of times it will be right when you take it, maybe for like five or 10 minutes and then it goes away. Some people also have allergic reactions. So at the injection site, you may get a little bit of a rash and again, that's more just from injecting, not necessarily from the peptide itself.
And then also headaches. Um, people have reported mild headaches, but, um, it's unclear whether SS 31 is a red cause. Again, and that just kind of one of the things you'll experience when taking peptides themselves. Um, now what about dosing? So this was hard to find. So I had to go to some insider people to fine this out, but, um, basically you can do it for 10 milligrams a day. When I would do, it is fasted first thing in the morning, and then not eat food for like 90 minutes after, especially to get the neuro. and cognitive boosting benefits that you're going to get from optimizing mitochondrial function.
So I would say faster in the morning and no food for at least 90 minutes to two hours after. And what I do is you can do it for eight weeks on and eight week off and just stay on that and see where you are after a year. A lot of times with these mitochondria peptides, you don't have to do them long because they work so well. have lasting effects because of the way they upregulate mitochondrial health is you don't have to stay on them all the time, but you're going to get benefits. So I would do it for eight weeks on a week soft, kind of see where you are. And if you want to keep going and you feel good, uh, continue that cycle for a year or so, you know, looking at.
three cycles per year doing it that way. That way it doesn't cost you too much and you're going to get good benefit. And you also going keep your receptors fresh to it and avoid the antibody buildup like you would get from other peptides where you using them too. So that's what I would do. Kind of see where are after a year. The 10 milligrams a day is enough to really get benefit and then a lot of people have reported A lot of the age-related or chronic disease they may be experiencing, you know, like fibromyalgia, MS, Hashimoto's, all these different things can kind of go away because of how it upregulates mitochondrial function. So that's pretty much the dosing schedule.
Again, there's not a lot out there on that. And then obviously you would inject this subcutaneously into the belly fat. just with an insulin needle. There are people that have gone up to like 50 milligrams a day if there's an acute condition. I've heard people with kidney disease using 50 milligrams a day and that like completely wiping out their kidney disease. So it is pretty powerful.
